Some news :
tested the fuel pump, no problem, good flow. Tried to make it run with the fuel pump removed from the head, the noise is still there.
After, I tried to pull spark plug caps, no rpm effect at idle when I pull off #3 & #4 caps
Yet spark at the end of the coil wires (spark 12mm long), spark with a spare plug inserted in the cap. Plugs 3 & 4 not wet, black coal color, blacker than the #1 & #2 cyl plugs.
So, a lot of things to check now, carbs slow and primary jets ?, perhaps a colortune test, change coils for dyna ones (what is the best place to buy it by the way ?).
Any advice welcome ;)

Checked the coils, HT resistance 14000 ohms ok, I cleaned the electrical connectors and it was the come-back of the #3 & 4 cylinder. But I think these coils may be weak.
To see if it's the case, I'd like to make it run with some HONDA 600 Hornet 03-06 coils : could these fit at your opinion ?

A little run this afternoon, a lot better concerning the smoothness of the engine !!!!
Still that knocking noise when accelerating from little throttle opening to mid-opening.
The last thing I have to explore (after putting brand new spark plugs) are the carbs : cyl. #3 is cold when I first start the bike, perhaps pilot jet on this carb a little clogged . . .

It's not uncommon for the engine's primary chain to rattle under load, especially if the carbs aren't synced properly, or out of tune. You may find it lessens after getting the carbs sorted.

+1 with Track T.
Just a small amount of dynamic synchronizing correction on my '75's carburetors greatly reduced several issues. tumb2

Thanks for sharing your experience !
Carb removal and inspection planned, to see if all is is order (they have been serviced by a professionnal before I buy it, but maybe a little clogged), and if it doesn't cure the noise, as ignition is OK now, engine stripping this winter ! :smash
To be continued . . . :-D

Wow. That's a lot of work you've done for the stand. With a little luck, you might still find that a motor teardown will not be necessary - but if it is, you certainly seem prepared to take that deep dive!

Professional carb builders work goes down the toilet if the tank and pump and associated tubing aren’t cleaned up. Tanks can harbor a lot of flaky rust at the top portion. Look with a mirror. Seems like the flakes breakdown into fine particles that get through the filter and screens and that 35 idle jet plugs quickly. From the looks of things, you’re well on it.
Whatever I suggest here should be given ample time for a moderator to delicately correct. I apologize in advance.

Checked the carbs : 2 idle jet a little clogged but not plugged, one float level a little low (23 instead of 21mm), no filters before the floats needles (hopefuly, can put ones from 1000 cbx ramps I had in stock).
After this little cleaning, very good idle (500 rpms I think !!!), but have to carry on, lack of battery/starter power (I didn't filled the carbs with fuel before starting, so the starter and perhaps coils heated . . . ).
To be continued . . .

In fact, the engine stopped cause it runned out of fuel . . . :-D (I made it run today and no coil problem, very good idle, althrought oise is still heardable when opening throttle).
The surprise of the day was to discover coolant fluid on the floor, under the right exhaust. After a few checks, coolant comes from the #3 cylinder (the area where the toctoc noise occured, and the cylinder that had misfires !!!). Perhaps a leaking head gasket is the solution to this problem . . .
